Prostaglandin-Induced Resorption of the Adult Rat Calvarium

چکیده
Prostaglandins are a family of naturally occurring fatty acids that participate in the inflammatory process' 4 and exhibit hormonelike effects.5,6 When prostaglandins are injected intradermally, they produce local inflammatory effects such as increased vascular permeability with edema and vasodilatation with erythema.7,8 They have been identified in inflammatory exudates9-11 and their biosynthesis is inhibited by nonsteroidal anti-inflammatory drugs.12-15 The prostaglandins PGE,, PGE2, and to a lesser extent PGF2a stimulate resorption of cultured fetal rat bones.16'17 In these systems E-type prostaglandins are more potent than is parathyroid hormone in producing bone resorption, although osteolytic action occurs at a slower rate. The action of the E-type prostaglandins and parathyroid hormone is accompanied by elevation of bone cyclic 3'-5' adenosine monophosphate.18 When they are injected systematically, prostaglandins are rapidly metabolized19'20 by 15 dehydrogenation and by reduction of the 13-14 double bond (Fig 1). The biologic activity of the metabolic products is reduced substantially. Because of the rapid inactivation and low

L-Carnitine potentiates the anti-inflammatory and antinociceptive effects of diclofenac sodium in an experimentally induced knee osteoarthritis rat model
Objective(s): The aim of the present research is to investigate the efficacy of L-carnitine (LC) as a complementary therapy to diclofenac sodium (Dic) treatment in a mono-iodoacetate (MIA) induced knee osteoarthritis (OA) rat model, with respect to pain relief and the underlying pathology.Materials and Methods: Fifty adult male albino ra...
متن کاملDirect extraction and assay of bone tissue collagenase and its relation to parathyroid-hormone-induced bone resorption.
A method has been developed for the quantitative extraction of collagenase from as little as one 19-day-fetal-mouse calvarium. About 20-40 munits of collagenase are extracted per mg of tissue, all in a latent form that, after proper activation, shows the typical properties of mammalian collagenase. Culturing the calvaria for 2 days with parathyroid hormone (PTH) increases their procollagenase c...
متن کاملEffects of DNA and prostaglandin synthesis inhibitors on the stimulation of bone resorption by epidermal growth factor in fetal rat long-bone cultures.
We examined two inhibitors of DNA synthesis, hydroxyurea (HU) and aphidicholin (APC), and two inhibitors of prostaglandin cyclooxygenase, indomethacin and flufenamic acid, for their effects on the resorptive responses of fetal rat long-bone cultures to epidermal growth factor (EGF) and parathyroid hormone (PTH). As we have previously found, HU decreased unstimulated 45Ca release but had little ...
متن کاملCrossing interaction of adrenergic, cholinergic, histaminergic and opioidergic systems on water intake in adult male rats
Several lines of evidence have indicated that many nuclei in the brain including preoptic nucleus, AV3V, subfornical organ, septal area and lateral hypothalamus are the targets of efferents from chemo-sensitive and pressure-sensitive systems. These areas may concern with regulation of fluid homeostasis. In the present study, intracerebroventricular injections were carried out in all experiments...
